چکیده

VER SINCE Paul Ehrlich’s discovery that granuleE laden cells (“mastzellen”) in the frog mesentery, originally observed by von Recklinghausen, possessed metachromatic staining properties,’” cells in the basophil and mast cell lineages have been the subject of much scrutiny. The description by dermatologists at the turn of the century of the condition known as urticaria pigmentosa, involving mast cell infiltration in the skin and other organs?4 and subsequent findings that mast cells contain various preformed mediators of inflammation such as histamine: led gradually to a widespread fascination with the role of these cells in biologic processes. In parallel with these observations were descriptions by Ehrlich’ and later by hematologists of the presence of a blood-borne metachromatic, histamine-producing granulocyte, the basophil, significantly prominent in a number of myeloproliferative diseases, especially in chronic myeloid leukemia: Historically, because of their common metachromatic staining properties, the basophil and the mast cell have been thought to be intimately related in terms of hematopoietic lineage, and the literature is indeed replete with this assumption. The mast cell in the tissue and the basophil in the blood have, however, not been proven to be derived from a common stem cell in either animals or humans; moreover, there appears to be an inverse relationship between the presence of basophils or mast cells as the prominent representative of a metachromatic cell among various species.’ This review will focus on the ontogeny of basophils and mast cells, exploring knowledge in this field as it relates to cytokine biology, allergy and inflammation, and clinical hematology. The extensive literature on the biology of the mast cell in various processes, such as wound healing, repair, and fibrosis or on basophil and mast cell secretory function, will be touched upon only briefly within the scope of this review.

Ex vivo Expansion and Differentiation of Mesenchymal Stem Cells from Goat Bone Marrow

Objective(s) Mesenchymal stem cells (MSCs) from large animals as goat which is genetically more closely related to human have rarely been gained attentions. The present study tried to isolate and characterize MSCs from goat bone marrow. Materials and Methods Fibroblastic cells appeared in goat marrow cell culture were expanded through several subcultures. Passaged-3 cells were then different...

Biological and Immunological Reactions in Parasitic Diseases

Processed antigen reacts selectively with lymphocytes bearing .appropriate surface immunoglobulin molecules resulting in the clonal proliferation of these cells, producing circulating immunoglobulin (humoral immunity) or non-circulating immunoglobulins (cellular immunity).  High levels of circulating immunoglobulins (lgM & lgG) occur in protozoa and helminths infections. Reaginic antibodies a...

Reduced mast cell and basophil numbers and function in Cpa3-Cre; Mcl-1fl/fl mice.

It has been reported that the intracellular antiapoptotic factor myeloid cell leukemia sequence 1 (Mcl-1) is required for mast cell survival in vitro, and that genetic manipulation of Mcl-1 can be used to delete individual hematopoietic cell populations in vivo. In the present study, we report the generation of C57BL/6 mice in which Cre recombinase is expressed under the control of a segment of...

Transcription factor IRF8 plays a critical role in the development of murine basophils and mast cells.

Basophils and mast cells play critical roles in host defense against pathogens and allergic disorders. However, the molecular mechanism by which these cells are generated is not completely understood. Here we demonstrate that interferon regulatory factor-8 (IRF8), a transcription factor essential for the development of several myeloid lineages, also regulates basophil and mast cell development....

Protein Profiling of the Secretome of FcεRI Activated RBL-2H3.1 Cells

Background: Secretory proteins of IgE receptor activated mast cells and basophils play a pivotal role in the generation of immediate and long term immune responses in allergy and type I hypersensitivity. Objective: The present study aims to generate a 2-D map and profile of proteins secreted from a high secretory variant of the rat basophilic leukemia cell line, RBL-2H3.1, which in view of the ...
